MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA Type-C) is a 16-amino acid mitochondrial-derived peptide encoded by the 12S rRNA region of the mitochondrial genome. Its primary target organ appears to be skeletal muscle, where its cellular actions inhibit the folate cycle and its tethered de novo purine biosynthesis, leading to AMPK activation — the master nutrient sensor involved in metabolic regulation. Under metabolic stress, MOTS-c translocates to the nucleus in an AMPK-dependent manner, where it binds to transcription factors and directs expression of nuclear genes to promote cellular homeostasis. Research has demonstrated that MOTS-c treatment prevented age-dependent and high-fat-diet-induced insulin resistance, as well as diet-induced obesity in mouse models. Exercise studies revealed an approximately 11.9-fold increase in endogenous MOTS-c levels in skeletal muscle following physical activity. MOTS-c has been shown to promote glucose utilization, enhance metabolic flexibility, inhibit oxidative stress, and improve mitochondrial function, positioning it as a significant peptide in metabolic, longevity, and exercise performance research.
